I study the mechanisms responsible for the instability of DNA repeats in various genomes. I am particularly interested in the mechanisms of expansions of triplet DNA repeats that cause numerous hereditary disorders in humans, including Fragile X mental retardation, Huntington's disease, myotonic dystrophy, and Friedreich's ataxia. My lab was the first to show that expansions could be due to the ability of these repeats to trick DNA replication into lengthening them. We are substantiating the replication model for repeat expansions in several experimental systems.
